One thing to keep in mind with humor is humor is about the mind being successfully tricked. To get humor in a speech, you create an expectation in the audience's mind, and then you go in a completely different direction from what they expected. A famous example is Henny Youngman's joke, "Take my wife...please!" The audience expects Henny to say, "for example", but his answer surprises them and so they find it humorous.
