To quickly get humor and comedy in your speech say something that gets your audience thinking one way while you go another way. For example, if people arrive late to your talk, and then seat themselves (or are seated) in the front row, you could say to them, "Can I get you something...like a watch?" The "can I get you something" sets up the expectation that your next words will be about providing some food or drink for the people. While your next "like a watch" changes direction and reminds everyone that these people are late and interrupting your show.
